An Overview of Our Hybridoma Development Service Schedule

Day -2 Prebleed each mouse.
Day 1 Immunize mice with antigen mixed with complete Freund's adjuvant .
Day 14 Immunize mice with antigen mixed with incomplete Freund's adjuvant.
Day 21 Collect immune bleeds.
Day 28 Immunize mice with antigen mixed with incomplete Freund's adjuvant.
Day 35 Collect immune bleeds.
Day 35 Send investigator 10 to 50 µl of each immune serum.
The investigator needs to have the immune serum analyzed during the next 2 days and have results ready to contribute to the consultation on Day 38.
Day 36-37 Our facility will independently monitor the immune bleeds for antigen-specific reactivities.
Day 38 Consult with the investigator and compare immune screening results. Decide which immune-responsive mice will be used for fusion(s).

Your first fusion will be scheduled in the first available date, usually with two weeks.

Starting from the day the fusion is done, the following is a representative schedule.

Day -3 Perform the fusion boost injection on a selected mouse.
Day 1 Perform hybridoma fusion technology
Day 14 Screen all hybridoma supernatants. Send investigator up to 20 putative positive supernatants for confirmation.*

The investigator needs to have these supernatants analyzed during the next 2 days and decide which of up to 4 hybridoma populations should be subcloned.

If no positives are identified we will schedule a second fusion.

Day 17 Subclone up to 4 positive hybridoma cell populations.
Day 35 Screen subcloned hybridoma supernatants. Send investigator up to 20 putative positive supernatants from subcloned hybridoma cell lines for confirmation.

The investigator needs to have these supernatants analyzed during the next 2 days and decide which 2 hybridoma cell lines should be used in a second round of subcloning.

Day 38 Second round subcloning on 2 of the hybridoma cell lines.
Day 52 Screen the twice subcloned hybridoma cell lines. Send investigator supernatants from twice subcloned hybridoma cell lines for confirmation. Expand cell lines.
Day 70 Send supernatants and frozen ampules containing twice subcloned hybridoma cell lines to the investigator and up to 20 ml hybriodma supernatant.

In most projects are completed within 4 to 6 months providing the first fusion was successful.

Mice are sacrificed at the conclusion of a successful project, unless client accepts mouse housing charges of $50/month for possible additional fusion(s).
